Embelin enhances the osteogenic potential of LPS-induced periodontal ligament stem cells by activating AMPK and SIRT1

Purpose: To investigate the role of embelin in periodontal ligament stem cells (PDLSCs) an vitro model periodontitis.Methods: Lipopolysaccharide (LPS)-stimulated PDLSCs was used to construct a periodontitis cell model. treatment group were pretreated with different concentrations Embelin, and CCK-8 TUNEL staining analyze viability apoptosis. Enzyme-linked immunosorbent assay (ELISA) kits evaluate levels inflammatory cytokines (TNF-α, IL-1β, IL-6, MCP-1) while reactive oxygen species assessed by 2',7'- dichlorodihydrofluorescein diacetate (DCFH-DA) staining. Subsequently, osteogenic marker, ALP activity protein expression Runx2, OCN BMP-2 evaluated western-blot assay; AMPK SIRT1 also determined using Western blot assay.Results: Embelin pretreatment inhibited apoptosis, factors, oxidative stress, but up-regulated ALP, OCN, (p < 0.05). In addition, phosphorylation regulated 0.05).Conclusion: exerts anti-inflammatory, anti-oxidative differentiation effects LPS-induced activating AMPK/SIRT1 signaling. Therefore, compound has potentials for use management periodontitis.
